تاثیر عصاره دانه سفالاریای سوری (Cephalaria syriaca L.)، تولید شده در تاریخ‌های کاشت و شرایط آبیاری مختلف بر کیفیت نوشیدنی کفیر

چکیده
سفالاریای سوری (Cephalaria syriaca L.) از جمله گیاهان دارویی یک­ساله می­باشد که تاکنون تحقیقات اندکی در مورد آن گزارش شده است. در پژوهش حاضر، ترکیبات فنولی و فلاونوئیدی کل، فعالیت ضد­اکسیدانی و ضد­رادیکالی سفالاریای سوری تحت شرایط اکولوژیک ارومیه (زمان­های کشت بهاره، انتظاری، پاییزه و رژیم آبیاری کامل، آبیاری تکمیلی، دیم) مورد مطالعه و مقایسه قرار گرفت. در این پژوهش، ابتدا 1 گرم از پودر دانه گیاهی با 10 میلی­لیتر حلال اتانول-آب (70:30) ترکیب سپس به مدت 3 ساعت بر روی شیک پلیت با دور 1000 rpm و در نهایت به مدت 15 دقیقه در دمای 40 درجه سانتی­گراد در حمام فرا صوت قرار داده شد. شناسایی و اندازه­گیری مقادیر فنولی تام و فلاونوئیدها توسط کروماتوگرافی مایع با کارایی بالا، و اندازه­گیری فعالیت ضد­اکسیدانی و ضد­رادیکالی توسط دستگاه اسپکتروفوتومتر اندازه­گیری شد. تجزیه و تحلیل نتایج با استفاده از نرم­افزار SAS و روش آزمون آنالیز واریانس انجام شد. نتایج نشان داد که عصاره استخراج شده مربوط به تیمار بدون آبیاری و کشت پاییزی، نسبت به دو رژیم دیگر آبیاری، افزایش معنی­داری در میزان فنول کل ( mg QUEg-1308/25)، فلاونوئید ( mg QUEg-1 202/2) و درصد مهار­کنندگی رادیکال DPPH (149/37)، درصد مهار رادیکال­های نیتریک اکسید (563/35) و سوپر اکسید (292/27) داشت. نتایج شناسایی ترکیبات فنولی نشان داد گالیک اسید، کلروژنیک اسید، کوماریک اسید و کروستین به­عنوان ترکیبات اصلی پلی­فنل سفالاریای سوری شناسایی شدند که این ترکیبات با افزایش تنش خشکی بر محتوای آنها در گیاه افزوده شد به طوری که در شرایط یکسان، گالیک اسید (mg QUEg-1 105952/1222) و سینامیک اسید (mg QUEg-1 86/0) به ترتیب بالاترین و کمترین مقدار را داشتند. نتایج ارگانولپتیک کفیر غنی­سازی شده نشان داد که تیمار 2/0 میلی­گرم پودر عصاره در 100 سی­سی کفیر بهترین نتایج را داشت. نتایج نهایی نشان داد می­توان از خواص فراسودمند گیاه سفالاریا سوری در محصولات دارویی و غذایی بهره­مند شد.

چکیده English

Cephalaria syriaca (Cephalaria syriaca L.) is one of the annual medicinal plants that little research has been reported about it. In the present study, the total phenolic and flavonoid compounds, antioxidant and antiradical activity of Cephalaria syria were studied and compared under the ecological conditions of Urmia (spring, waiting, autumn planting times and full irrigation regime, supplementary irrigation, dryland). In this research, first, 1 gram of plant powder was mixed with 10 ml of ethanol-water solvent (70:30), then mixed for 3 hours on a shaker plate at 1000 rpm and finally for 15 minutes at a temperature of 40 degrees Celsius in a bath. An ultrasound was performed. Total phenolics and flavonoids were identified and measured by high-performance liquid chromatography, and antioxidant and antiradical activity was measured by spectrophotometer. The results were analyzed using SAS software and analysis of variance test method. The results showed that the extracted extract related to the treatment without irrigation and autumn cultivation, compared to the other two irrigation regimes, showed a significant increase in the amount of total phenol (125.308 mg QUEg), flavonoid (12.202 mg QUEg) and DPPH radical inhibition percentage. (37.149), inhibition percentage of nitric oxide radicals (35.563) and superoxide (27.292). The results of the identification of phenolic compounds showed that gallic acid, chlorogenic acid, coumaric acid and crostin were identified as the main polyphenol compounds of Cephalaria syria, and these compounds increased their content in the plant with the increase of drought stress, so that under the same conditions, gallic acid (mg QUEg -1 1222.105952) and cinnamic acid (mg QUEg-1 0.86) had the highest and lowest values, respectively. The organoleptic results of enriched kefir showed that the treatment of 0.2 mg of extract powder in 100 cc of kefir had the best results.
