بررسی اثر حلال و روش استخراج بر محتوی فیتوشیمیایی و فعالیت آنتی‌اکسیدانی عصاره برگ مورینگا الیفرا (Moringa oleifera)

چکیده
گیاهان منبع غنی از ترکیبات فنلی هستند که مهم‌ترین آنتی‌اکسیدان‌های طبیعی به شمار می‌روند. هدف از این تحقیق بررسی تأثیر روش‌های استخراج و اثرحلال های مختلف بر راندمان استخراج، محتوی فنل کل، فلاونوئید کل و فعالیت آنتی اکسیدانی عصاره برگ گیاه مورینگا الیفرا بود. بدین منظور برگ‌ها بعد از خشکاندن آسیاب شد و عصاره با استفاده از حلال‌های آب مقطر، استون و متانول با روش‌های خیساندن، سوکسله و امواج غیرمستقیم اولتراسونیک با فرکانس 70 کیلوهرتز استخراج شد. محتوی کل ترکیبات فنلی با روش فولین –سیوکالتیو و محتوی تام ترکیبات فلاونوئیدی با روش رنگ سنجی کلرید آلومینیوم اندازه‌گیری شد. برای اندازه‌گیری فعالیت آنتی‌اکسیدانی عصاره‌ها از روش مهار رادیکال‌های آزاد 2 و 2 دی فنیل 1-پیکریل هیدرازیل (DPPH) استفاده شد. فعالیت آنتی‌اکسیدانی عصاره‌ها با اسید آسکوربیک مقایسه گردید. بالاترین راندمان استخراج مربوط به عصاره استحصال شده با حلال استون و روش استخراج سوکسله (0.34±24.40 درصد) بود. متانول در روش استخراج سوکسله بالاترین تاثیر را بر محتوی فنل کل (1.35±24.79 میلی گرم گالیک اسید در گرم عصاره خشک) و فلاونوئید کل (1.45±81.14 میلی‌گرم کوئرستین در گرم عصاره خشک) نشان داد. در همه نمونه‌ها با افزایش غلظت عصاره، میزان مهار رادیکال‌های آزاد DPPH افزایش یافت و بالاترین قدرت حذف رادیکال‌های آزاد در عصاره متانولی حاصل از روش استخراج سوکسله (0.65±49.975=50 IC میکروگرم در میلی‌لیتر) مشاهده شد. در نهایت می توان نتیجه گرفت متانول بهترین حلال و استخراج با سوکسله بهترین روش برای استخراج ترکیبات فنلی و فلاونوئیدی از برگ مورینگا الیفرا می باشد.

Plants are rich source of phenolic compounds, which are the most important natural antioxidants. The aim of this study was to investigate the effect of extraction methods and different solvents on extraction yield, total phenolic content (TPC), total flavonoid content (TFC) and free radicals scavenging activity of DPPH in Moringa oleifera leaf extract. For this, the leaves were grinding after dried followed by different extraction techniques like soaking, Soxhlet and ultrasound (Frequency: 70 kHz) using distilled water, acetone and methanol as solvent. TPC and TFC were measured using Folin–Ciocalteu method and colorimetric assay. Antioxidant activity of resulted extracts was measured using 2, 2-Diphenyl-1-picryl-hydrazyl (DPPH). The antioxidant activity of the extracts was compared with ascorbic acid. The highest extraction efficiency was obtained in Soxhlet method use acetone as solvent (24.40±0.34%). Methanol in Soxhlet extraction method showed the highest effect on TPC (24.79±1.35 mg GAE/g dry sample) and TFC (81.14±1.45 mg QU/g dry sample). In all samples, increased DPPH free radical scavenging when increased concentration of the extracts. The highest radical scavenging was observed in methanol extracts obtained by Soxhlet (IC50=49.97±0.65). Finally, it could be concluded that, methanol was the best solvent and Soxhlet was the best method for extraction of phenolic and flavonoid compounds in in the leaves of Moringa oleifera.
