ارزیابی حداقل غلظت مهارکنندگی و کشندگی عصاره‌های پنیرک (Malva sylvestris) و بر همکنش آن‌ها بر تعدادی از باکتری‌های بیماری‌زا در شرایط آزمایشگاهی

چکیده
گیاه پنیرک با نام علمی Malva sylvestris متعلق به خانواده پنیرکیان است. ترکیبات مؤثر و فعال موجود در گیاه پنیرک می‏تواند دلیلی برای فعالیت زیستی گیاه و در نهایت کاربردهای درمانی باشد. هدف از این پژوهش ارزیابی اثر ضد میکروبی گیاه پنیرک بر تعدادی از باکتری‏های عامل بیماری‏های عفونی بود. عصاره‏گیری از گیاه با روش خیساندن (ماسراسیون) انجام شد. برای سنجش کیفی اثر ضد میکروبی گیاه پنیرک از دو روش دیسک دیفیوژن و تمام ظرف استفاده گردید. برای ارزیابی کمی فعالیت ضد میکروبی نیز از روش میکرودایلوشن براث و روش بازدارنده افتراقی (برهمکنش) استفاده شد. حداقل غلظت‏کشندگی نیز به روش پورپلیت تعیین گردید. نتایج نشان داد، غلظت 100 میلی‏گرم بر میلی‏لیتر عصاره اتانولی بر باکتری‏های استرپتوکوکوس پیوژنز و استافیلوکوکوس اورئوس بیشترین اثر را داشت، به طوری که اختلاف معنی‏داری با تمامی تیمارها ی دیگر از خود نشان داد. انتروباکتر ائروژینوزا بیشترین مقاومت به عصاره‏های آبی و اتانولی گیاه پنیرک داشت. حداقل غلظت بازدارندگی عصاره اتانولی برای باکتری­های استرپتوکوکوس پیوژنز، استافیلوکوکوس اورئوس، اشرشیا کلی و انتروباکتر ائروژینوزا به ترتیب 8، 8، 64 و 64 میلی­گرم بر میلی‌لیتر بود. در حالی که برای عصاره آبی به ترتیب 16، 32، 128 و 128 میلی‏گرم بر میلی لیتر بود. در نهایت می‏توان گفت عصاره اتانولی گیاه پنیرک در مقایسه با عصاره آبی اثر بازدارندگی بیشتری بر سویه‏های بیماری‏زا داشت. همچنین نتایج نشان داد، عصاره‏های آبی و اتانولی گیاه پنیرک بر باکتری‏های گرم مثبت نسبت به باکتری های گرم منفی اثر ضد میکروبی بیشتری نشان دادند.

Investigation of the minimum inhibitory concentration and minimum bactericidal concentration Malva sylvestris extracts and their interaction on some pathogenic bacteria in vitro

Malva sylvestris belongs to Malvaceae familyThe biological active compounds of Malva sylvestris describe probably it’s biological activities and therapeutic activity. The purpose of this study was to investigate the antibacterial effect of Malva sylvestris on some pathogenic bacteria “in vitro”.In this study, the extraction was carried out by the maceration method. The pour plate method and disk diffusion method were adopted to determine the susceptibility of the pathogenic strains against Malva sylvestris extracts. The broth microdilution and fractional inhibitory concentration index (FICI) were employed to determine antimicrobial effect. The minimum bactericidal concentration was measured by pour plate method. The concentration 100 mg/ml of the ethanolic extract showed the best result on Streptococcus pyogenes and Staphylococcus aureus (p<0.05). Enterobacter aeruginosa was resistant to most of the aqueous and ethanolic Malva sylvestris extracts. Minimum inhibitory concentration of the ethanolic extract of Malva sylvestris for Streptococcus pyogenes, Staphylococcus aureus, Escherichia coli and Enterobacter aeruginosa were 8, 8, 64 and 64 mg/ml, respectively and the aqueous extract were 16, 32, 128, and 128 mg/ml, respectively.The ethanolic extract of Malva sylvestris had more inhibitory effect than the aqueous extracts on pathogenic strains. Furthermore, aqueous and ethanolic Malva sylvestris extracts showed greater inhibitory effect on gram-positive bacteria in comparison with gram-negative bacteria.
