مقایسه میزان فعالیت آنتی اکسیدانی در دو گیاه ریحان (Ocimum basilicum)و گل حنا (Impatiens walleriana) و بررسی خواص ضد میکروبی و ضد سرطانی آنها بر روی سرطان تخمدان

چکیده
سرطان تخمدان یکی از کشنده ترین سرطان ها در خانم ها محسوب می شود و باعث ایجاد سلول های غیرطبیعی سرطانی میشود که این سلول ها توانایی گسترش و حمله به سایر قسمت های بدن را دارا میباشد. متابولیت های ثانویه از لحاظ ساختاری و شیمیایی بسیار متنوع می باشند و نقش مهمی در درمان انواع سرطان ایفا می کنند. در این مطالعه عصاره دو گیاه تحت تأثیر سه حلال ( آب ، اتانول 80 % و متانول 80 % ) روش عصاره گیری خیساندن گرفته شد و با استفاده از روش فولین سیوکالتو ، میزان ترکیبات فنلی و با روش های DPPH (دی فنیل پیکریل هیدرازیل) و FRAP ( توانایی احیای آهن ) فعالیت آنتی اکسیدانی دو گیاه سنجیده شد. سپس از بین حلال ها، حلال آب که دارای بیشترین خاصیت آنتی اکسیدانی بود جهت بررسی خواص ضد سرطانی بر روی سلول های سرطان تخمدان انتخاب گردید که کشنده ترین عصاره مربوط به عصاره آبی گیاه ریحان در 72 ساعت با میزان IC50 برابر با 001/0 ± 105/1 میلی گرم بر میلی لیتر بوده است. میزان ترکیبات فنلی در دو گیاه تحت تأثیر حلال متانول 80 % بیشترین میزان بود و بیشترین فعالیت آنتی اکسیدانی نیز در دو گیاه در حلال آب مشاهده شد . در بررسی خاصیت ضد میکروبی به روش انتشار دیسکی ، این خاصیت فقط در عصاره های اتانولی دو گیاه دیده شد. در سنجش MIC (Minimum Inhibitory Concentration) و(Minimum Bactericidal Concentration) MBC عصاره های اتانولی و متانولی گیاه ریحان و در گل حنا در عصاره های آبی و اتانولی دارای بیشترین اثر بازدارندگی و همچنین کشندگی بودند.

Abstract

Ovarian cancer is one of the deadliest cancers in women causing abnormal cancerous cells that can spread and metastasize to other parts of the body. Secondary metabolites are highly variable in terms of structural and chemical properties and they are very important for plants survival. In this study, the extracts of two plants using three different solvents (water, ethanol 80% and methanol 80%) and water bath extraction method were obtained. phenolic compounds of extracts were determined by Folin Sioculto method. DPPH (2, 2-Diphenyl-1-picrylhydrazyl) and FRAP (Ferric reducing ability of plasma) methods were performed for determination of antioxidant properties. The amount of phenolic compounds in two plants was higher in 80% methanol solvent and the most antioxidant activities were observed in water solvent of extracts. Water solvent had the highest antioxidant activity and was selected to investigate the anti-cancer properties of extracts on ovarian cancer cells. Water extract of Ocimum basilicum within72 hours had the highest cytotoxic effect with an IC50 value of 1.105 ± 0.001 mg / ml. In study of antimicrobial activity by disc diffusion method, the highest antibacterial properties were only in ethanol extracts of two plants. In the measurement of MIC and MBC, ethanol and methanol extracts of Ocimum basilicum and water and ethanol extracts in Imatiens walleriana had the most inhibitory and bactericide effects.
