تحلیل ویژگی‌های عملکردی پروتئین لوبیا سیاه با تأکید بر خواص حلالیت، کف‌سازی و پایداری آن

چکیده
در چشم‌انداز رو به رشد صنایع غذایی، توجه فزاینده به غذاهایی با ساختار کف به عنوان مولفه‌هایی سالم و جذاب، ضرورت درک عمیق‌تر خواص عملکردی پروتئین‌های گیاهی را ایجاب می‌کند. پروتئین‌های گیاهی، به‌ویژه پروتئین‌های لوبیای سیاه(Phaseolus vulgaris) ، به عنوان جایگزینی سالم و طبیعی برای پروتئین‌های حیوانی، مورد توجه قرار گرفته‌اند. با این حال، شناخت عمیق‌تر از پارامترهای محیطی مؤثر بر خواص حلالیت، کف‌کنندگی و پایداری این پروتئین‌ها، نقشی کلیدی در بهبود کارایی آنها ایفا می‌کند. در این مطالعه، تاثیر شرایط فیزیکوشیمیایی شامل pH قلیایی(11-9)، pH ایزوالکتریک (5-4) و دما (24-4 درجه سانتی‌گراد) بر میزان حلالیت، ظرفیت تشکیل و پایداری کف پروتئین‌های لوبیای سیاه مورد بررسی قرار گرفت. نتایج نشان دادند که افزایش pH قلیایی و دما، به بهبود چشمگیر در ساختار پروتئین و تشکیل لایه‌ای پایدارتر منجر شده است. دماهای بالاتر سبب دناتوراسیون جزئی و ظهور مناطق آبگریز شد که شبکه‌های پروتئینی قوی‌تر ایجاد می‌کند. در شرایط بهینه فیزیکوشیمیایی شامل pH قلیایی(10.08)، pH ایزوالکتریک (4) و دما (24 درجه سانتی‌گراد)، بیشترین میزان حلالیت (1.16%)، بیشترین ظرفیت کف کنندگی (253.02%) و بیشترین پایداری کف در 30 دقیقه (111.09%) و در 60 دقیقه (57.80%) مشاهده شد. نتایج پیش‌بینی نرم افزار با داده‌های تجربی تفاوت معنی‌داری نداشت (p>0.05). این یافته‌ها نه تنها درک بیشتری از مکانیسم‌های تشکیل و پایداری کف ارائه می‌دهد، بلکه مسیر را برای توسعه محصولات فومی گیاهی با خواص فیزیکی و عملکردی بهینه هموار می‌سازد.

چکیده English

In the growing food industry landscape, the increasing attention to foam-structured foods as healthy and attractive components necessitates a deeper understanding of the functional properties of plant proteins. Plant proteins, especially black bean (Phaseolus vulgaris) proteins, have been considered as healthy and natural alternatives to animal proteins. However, a deep understanding of the environmental parameters affecting the solubility, foaming and stability properties of these proteins plays a key role in improving their performance. In this study, the effects of physicochemical conditions including alkaline pH (9-11), isoelectric pH (4-5) and temperature (4-24 °C) on the solubility, foaming capacity and stability of black bean proteins were investigated. The results showed that increasing alkaline pH and temperature led to a significant improvement in protein structure and the formation of a more stable layer. Higher temperatures caused partial denaturation and the emergence of hydrophobic regions, which create stronger protein networks. Under the optimal physicochemical conditions including alkaline pH (10.08), isoelectric pH (4) and temperature (24°C), the highest solubility (1.15%), highest foaming capacity (253.02%), and highest foam stability at 30 min (111.09%) and 60 min (57.80%) were observed. The software prediction results did not differ significantly from the experimental data (p>0.05). These findings not only provide a greater understanding of the mechanisms of foam formation and stability, but also pave the way for the development of plant-based foam products with optimized physical and functional properties.
