بهینه ‌سازی شرایط استخراج ترکیبات زیست فعال از گلبرگ های گل ‌راعی به روش سطح پاسخ

چکیده
در این پژوهش جهت عصاره ‌گیری از گلهای گل‌ راعی از امواج فراصوت استفاده شد. اثر نسبت متانول به آب (77/24، 35، 50، 65 و 22/75 درصد)، زمان (59/1، 5، 10، 15 و 40/18 دقیقه) و دما (77/24، 35، 50، 65 و 22/75 درجه‌ سانتی ‌گراد) بر استخراج ترکیبات فنولی، فلاونوئیدی و فعالیت آنتی ‌اکسیدانی عصاره به دو روش قدرت آنتی اکسیدانی احیاء آهن (FRAP) و دی ‌فنیل پیکریل هیدرازیل (%DPPH) با استفاده از روش سطح پاسخ و طرح مرکب مرکزی بررسی شد. بر اساس نتایج بهترین نسبت تیمارها مربوط به نسبت 50 درصد متانول به آب، زمان 15 دقیقه و دمای 70 درجه سانتیگراد بود. در شرایط بهینه مقدار فنول کل 46/28 میلیگرم گالیک‌‌ اسید بر گرم وزن خشک، مقدار فلاونوئید کل 75/19 میلی ‌گرم کوئرسیتین بر گرم وزن خشک، مقدار فعالیت آنتی‌ اکسیدانی (%DPPH) 23/89 درصد و مقدار فعالیت آنتی ‌اکسیدانی (FRAP) 57/3 میکرومول آهن در گرم وزن خشک بود. مطلوبیت بدست آمده در شرایط بهینه برای متغیرها و پاسخ ‌های مورد بررسی 895/0 بود. بر طبق نتایج ترکیبی از آب و متانول نسبت به استفاده انفرادی هر کدام از حلالها در استخراج پلی فنول ها کاراتر عمل کرده است. از سویی دیگر افزایش دما (حدود 15 دقیقه) و زمان (حدود 65 درجه سانتی ‌گراد) عصاره‌ گیری، منجر به افزایش ترکیبات فنولی می شود. با توجه به کارایی امواج فراصوت در بهبود فرآیند استخراج نسبت به سایر روش ‌های عصاره‌ گیری و تطابق داده ‌های حاصل از آزمایش با مدل ‌های پیش ‌‌بینی شده توسط روش سطح پاسخ، به نظر می‌ رسد که تلفیق این دو روش در بهینه ‌سازی استخراج آنتی ‌اکسیدان‌ های فنولی از گل راعی بسیار مناسب است.

In the present study, the use of the ultrasonic apparatus has been investigated in the extraction process of Hypericum perforatum L. flowers. The effect of methanol to water ratio (24.77, 35, 50, 65, and 75.22 percent), time (1.59, 5, 10, 15, and 18.40 minutes), and temperature (24.77, 35, 50, 65, and 75.22 °C) on the extraction of phenolic compounds, flavonoids and antioxidant activity of the extract using two methods of 2,2-diphenyl-1-picrylhydrazyl (DPPH%) and (FRAP) Ferric Reducing antioxidant power (FRAP) were studied using the response surface method and central composite design. The results of RSM showed that the highest extraction efficiency was obtained at 50% methanol/water ratio, 70 °C, and 15 min. Under optimal conditions, the corresponding values for total phenol content, total flavonoid content, and antioxidant assays of %DPPH and FRAP were 28.46 (mg gallic acid equivalent (GAE) g-1 dry weight (DW)), 19.75 (mg quercetin g−1 DW), 89.23 (%DPPH), and 3.57 FRAP µmol Fe++ /g DW respectively. In addition, results express the effectiveness of the proposed conditions and reliability of central composite design analysis on the extraction of phenolic antioxidants from H. perforatum flowers. Using ultrasonic is a low-cost, simple, and efficient method for the extraction of phenolic antioxidants, which increases the extraction efficiency and thermal protection of phenolic antioxidants.
