تاثیر نوع حلال و شرایط استخراج بر عملکرد استخراج، محتوای ترکیبات فنولی کل و فعالیت ضد اکسایشی عصاره حاوی ترکیبات زیست فعال از اندام هوایی گیاه همیشه بهار (Calendula officinalis L.)

چکیده
در مطالعه حاضر عصاره حاوی ترکیبات زیست فعال از اندام هوایی گیاه دارویی همیشه بهار (Calendula officinalis L.) به روش خیساندن با استفاده از حلال­های آب، اتانول، استون، مخلوط اتانول/آب و استون/آب استخراج گردید. در میان حلال­های مورد مطالعه عصاره اتانولی حاوی بالاترین ترکیبات فنولی کل بود. تاثیر نسبت حلال به ماده اولیه (40:1-10:1 میلی­لیتر بر گرم) و اندازه ذرات (36/2-5/0 میلی­متر) بر عملکرد کمی و کیفی استخراج عصاره اتانولی در دمای محیط (25 درجه سلسیوس) نیز مورد بررسی قرار گرفت. بر اساس نتایج به دست آمده با استفاده از نسبت 30:1 میلی­لیتر بر گرم ماده اولیه و اندازه ذراتی معادل 1 میلی­متر در دمای محیط می­توان به بالاترین عملکرد کمی استخراج عصاره خام و ترکیبات فنولی کل به ترتیب معادل 17/0±5/3 درصد و 08/1±94/36 میلی­گرم معادل گالیک اسید در گرم نمونه دست یافت. فعالیت ضد اکسایشی عصاره به دست آمده تحت شرایط فوق در غلظت 1 میلی­گرم بر میلی­لیتر به ترتیب معادل 68/2±95/62 درصد مهار رادیکال­های آزاد 1،1- دی فنیل-2- پیکریل- هیدرازیل (DPPH) و 20/1±18/153 میکرومول یون آهن در لیتر برای قدرت احیا کنندگی آهن (FRAP) بود. ضریب همبستگی بالا (9594/0<R2) بین میزان ترکیبات فنولی کل و فعالیت ضد اکسایشی حاکی از نقش کلیدی ترکیبات فنولی در بروز فعالیت ضد اکسایشی می­باشد. در نهایت عصاره اتانولی به دست آمده از اندام هوایی همیشه بهار تحت شرایط مذکور را می­توان به عنوان یک منبع طبیعی از ترکیبات ضد اکسایش فنولی جهت استفاده در صنایع غذایی، دارویی به عنوان نگهدارنده طبیعی معرفی نمود.

In the present study, the extract containing bioactive compounds from marigold (Calendula officinalis L.) medicinal plant was extracted using water, ethanol, acetone, ethanol/water and acetone/water as solvent. Among the solvents studied, the ethanolic extract contained the highest total phenolic compounds. The effect of the solvent to raw material ratio (40:1-10:1 mL/g) and particle size (0.5-2.36 mm) on the quantitative and qualitative properties of ethanolic extract at ambient temperature (25°C) were investigated. Based on the results, the highest crude extraction yield and total phenolic compounds equal to 3.5 ± 0.17% and 36.94 ± 0.81 mg GAE/gram of sample was obtained using a ratio of 30:1 mL/g and a particle size of 1 mm at ambient temperature. At the concentration of 1 mg/ml, the antioxidant activity of the extract was 62.95 ± 2.68 % and 153.18 ± 1.20 μΜ Fe2+/L, respectively. The high correlation coefficient (R2>09594) between total phenolic compounds and antioxidant activity indicates the key role of phenolic compounds in antioxidant activity. Finally, the ethanolic extract obtained from C. officinalis under mentioned conditions can be introduced as a natural source of phenolic antioxidants for application in the food and nutraceutical industry as a natural preservative.
