بررسی و تعیین حداقل غلظت بازدارندگی(MIC) دی استات سدیم بر برخی از میکروارگانیسم های عامل فساد در نوشابه های گازدار در محیط کشت
Determination of minimum inhibitory concentration (MIC) of sodium diacetate on spoilage microorganisms in carbonated beverages by culture media
چکیده
دی استات سدیم از گروه مواد نگهدارنده شیمیایی و ایمن بوده و به عنوان بازدارنده کپک و مخمر و برخی باکتری هامعرفی شده است. .در این تحقیق اثر ضد میکروبی دی استات سدیم در جلوگیری از رشد برخی از میکروارگانیسم های عامل فساد نوشابه های گاز دار در محیط کشت با استفاده از روش حساسیت رقت های مایع مورد بررسی قرار گرفته است. در مورد مخمر <em>Saccharomycess cerevisiae</em> در تعداد 10، 10<sup>2</sup>، 10<sup>3</sup>،10<sup>5 </sup>مخمر در هر میلی لیتربه ترتیب غلظت های <sup> </sup>ppm 5000،1250،313،156 دی استات سدیم به عنوان MIC شناخته شد.در مورد مخمر <em>Candida krusei</em> در تعداد10، 10<sup>2</sup>، 10<sup>3</sup>،10<sup>5 </sup>مخمردر هر میلی لیتر به ترتیب غلظت های ppm<sup> </sup>5000,2500,1250,625 دی استات سدیم به عنوان MIC شناخته شد. در مورد باکتری <em>Leuconostoc mesenteroides</em> در تعداد 10<sup>2</sup> ،10<sup>4</sup>باکتری در هر میلی لیتربه ترتیب غلظت های ppm5000,2500 دی استات سدیم به عنوان MIC شناخته شد. در مورد باکتری<em> Lactobacillus delbrukii</em>درتعداد10<sup>2</sup>باکتری در هر میلی لیتر غلظت ppm2500 دی استات سدیم به عنوان MIC شناخته شد. در مورد کپک<em> Aspergillus</em> <em>niger</em> در کلیه رقت های تهیه شده از سوسپانسیون هاگ کپک ودر حضورغلظت های مختلف از دی استات سدیم رشد مشاهده شد. در نتیجه هیچ غلظتی به عنوانMIC انتخاب نشد. نتایج نشان داد که دی استات سدیم دارای خواص بازدارندگی برروی مخمرها و باکتری های مورد بررسی بوده و فاقد تاثیر بازدارندگی بر روی کپک <em>Aspergillus</em> <em>niger</em><em> </em>است.
Sodium diacetate is a safe chemical presevative which is used as an inhibitor against mould, yeast and some bacteria. In this study anti-microbial effect of sodium diacetate on preventing the growth of some spoilage microorganisms in carbonated beverages was investigated by broth diulation suceptibility test in medium. At the concentrations of 10<sup>1</sup>, 10<sup>2</sup>, 10<sup>3</sup> and 10<sup>5</sup> cells of <em>Saccharomycess cerevisiae</em> per ml 156, 313, 1250 and 5000 ppm of sodium diacetate were respectively determined as the minimum inhibitory concentrations (MIC) preventing growth of the yeast. As for<em> Candida krusei</em>, 625, 1250, 2500 and 5000 ppm of sodium diacetate were MIC inhibiting the growth of the yeast at concentrations of respectively 10<sup>1</sup>, 10<sup>2</sup>, 10<sup>3</sup> and 10<sup>5 </sup>cells/ml. 2500 and 5000 ppm of sodium diacetate were determined as MIC inhibiting the growth of <em>Leuconostac mesenteroides</em> at the concentrations of respectively 10<sup>2</sup>, 10<sup>4</sup> bacteria/ml, and for 10<sup>2</sup> bacteria/ml of <em>Lactobacillus delbrukii</em> was prevented by adding 2500 ppm sodium diacetate. No inhibitory effects of different concentrations of sodium diacetate observed at none of the prepared spore suspension of <em>Aspergilus niger, </em>so no concentrations of sodium diacetate were determined as MIC for this species of mould.The results show that sodium diacetate has inhibitory effects on the above selected yeast and bacteria but no inhibitory effects on the mould of <em>Aspergilus niger.</em>
